Growing cultures to mid-log

if you plan to do gene expression studies (like realtime PCR or microarray) with the RNA, you'll want to make sure that your culture (or both cultures, if you're comparing two or more conditions) are at the same growth phase, since this can wildly alter the expression of many genes.
